A green one-pot Biginelli synthesis of 3,4 ..

Various Biginelli compounds (dihydropyrimidinones) have been synthesized efficiently and in high yields under mild, solvent-free, and eco-friendly conditions in a one-pot reaction of 1,3-dicarbonyl compounds, aldehydes, and urea/thiourea/acetyl thiourea using lithium-acetate as a novel catalyst without the addition of any proton source. Comparative catalytic efficiency of lithium-acetate and polyphosphoric acid to catalyze Biginelli condensation is also studied under neat conditions. The reaction is carried out in the absence of any solvent and represents an improvement of the classical Biginelli protocol and an advantage in comparison with FeCl3·6H2O, NiCl2·6H2O and CoCl2·6H2O that were used with HCl as a cocatalyst. Compared to classical Biginelli reaction conditions, the present method has advantages of good yields, short reaction times, and experimental simplicity. The obtained products have been identified by spectral (1H NMR and IR) data and their melting points. The prepared compounds are evaluated for anticancer activity against two human cancer cell lines (lung cancer cell line A549 and breast cancer cell line MCF7).

ABSTRACT: Novel Schiff base comprising 4-aminoantipyrine with Biginelli adducts of salicylaldehyde has been designed and synthesised. Solid metal complexes of this Schiff base with Ni(II),Zn(II),Cd(II) and Hg(II) metal ions were synthesised and characterised by elemental analysis, magnetic susceptibility, molar conductance, ESI mass, IR,NMR and UV-Vis spectral studies. From the data it has been observed that the complexes had the composition of ML2 type, diamagnetic nature and a tetrahedral geometry around the metal ion except Ni(II) complex which has square planar geometry. The Ligand and its complexes have been screened for their in vitro antifungal and in vitro antibacterial activities against the fungi i.e., Candida albicans, Aspergillus flavus, Pencillium spp., Aspergillus niger and Trichophyton and the gram-negative bacteria Escherichia coli, Vibrio spp., Pseudomonas aeroginosa, Vibrio parahaemolytics, Salmonella spp., Aeromonas spp., Klebsiella spp., Proteus spp. and gram-positive bacteria Bacillus spp and Staphylococcus aureus. The in vitro antimicrobial activities of these metal complexes found to be stronger than the ligand.


A thorough search of literature reveals that no work has been done on the transition metal complexes of Schiff base condensed by 4-aminoantipyrine with Biginelli reaction through salicylaldehyde which is a characteristic aroma component of buck wheat 12 and have good antimicrobial activity against certain bacteria or fungi 13. Moreover Biginelli reactions (Multi component reactions) are of increasing importance in the organic and medicinal chemistry due to their high degree of atom economy, applications in the combinatorial chemistry and diversity oriented synthesis 14.

In present context, bidentate ligand from condensing 4-amionoantpyrine with Biginelliadduct of salicylaldehyde was synthesised. The synthesised compound and its chelation behaviour with metal ions were studied through physical parameters and spectral analysis. The in vitro antimicrobial properties of ligand and metal complexes were also investigated.

Perusal of literature revealed that the Biginelli reaction, which was discovered more than a century ago, is one of the most important MCRs for the synthesis of dihydropyrimidinones based on acid-catalyzed three component condensation of β-dicarbonyl compound, an aldehyde, and urea or thiourea [, ].
